How to steps
1: Write down the height and with of your map in pixels.
2: Determine how many days it would take for your adventurer to walk from the top to the bottom or form the right to the left of the map.
3. A human can walk, if they are in reasonably good shape 20 miles a day. The Roman legions in full armor could walk 18 miles in one day. These were professional soldiers.
4. Take your Travel days X it by 20 (days x 20) this will get your scale. Adjust if you want to meet a per inch / per x miles.
5. Grab google earth, measure a section of the world to gain an understanding of real world scale, and also to find how big your biomes are.

Numbers used: human can walk 20 miles in a day. A fully armed solider 18 miles in a day.
Biomes are roughly 1000 miles to 500 miles north to south.
